Divyendu Sharma began his career with a minor role as a goon in Aaja Nachle (2007) but struggled to find consistent work over the next few years. His breakthrough came in 2011 with the comedy Pyaar Ka Punchnama, where he played the quirky supporting character Liquid. The film, a sleeper hit, earned him the Screen Award for Most Promising Newcomer – Male and marked the official launch of his career.

In 2013, Divyendu Sharma appeared as the poet Omi in David Dhawan’s remake of Chashme Baddoor, another commercially successful project. He followed this with supporting roles in films such as Ekkees Toppon Ki Salaami (2014) alongside Anupam Kher and his first lead in Dilliwali Zaalim Girlfriend (2015) opposite Prachi Mishra, though both had limited impact on his career trajectory.

Following Mirzapur, Divyendu Sharma appeared in films such as Badnaam Gali (2019), Kanpuriye (2019), Shukranu (2020), Mere Desh Ki Dharti (2022), Odd Couple (2022), and Thai Massage (2023), along with web series like Bicchoo Ka Khel (2020) and Salt City (2022). In 2023, he played Balwant Yadav, a heroic yet morally complex character, in Netflix’s miniseries The Railway Men, based on the 1984 Bhopal disaster.

In 2024, Divyendu starred in Madgaon Express and the Disney+ comedy-drama series Life Hill Gayi, where he portrayed a privileged young man forced to manage a village hotel alongside his sister. While the projects received mixed to negative reviews, Divyendu continues to solidify his reputation as a versatile and compelling actor in both films and digital media.
